GFAP+ and PLP1+EGCs complement each other to maintain intestinal homeostasis.③ GFAP+EGCs specific amplification regulates LCR5+ ISC, stimulates ISC activity, promotes early mucosal regeneration, and repairs intestinal injury;④ The mechanism of GFAP+EGCs in injury repair of IBD patients is conservative. During enteritis repair, GFAP+EGCs activates WNT signaling pathway in ISC and promotes early intestinal regeneration.⑤ DSS mouse experiment showed that GFAP+EGCs provided key WNT signal for colitis repair.The intestinal nervous system consists of a rich network of intestinal glial cells (EGCs) and neurons, and although EGCs are adjacent to intestinal stem cells, their effect on stem cells is unknown.EGCs control Stem Cell activity during intestinal homeostasis and regeneration.Single-cell analysis and mouse genetic studies have shown that specific populations of GFAP+EGCs are activated during the early stages of repair and chronic inflammation and stimulate stem cell responses that promote early mucosal regeneration.This study highlights the mechanisms by which functional glial heterogeneity affects intestinal stem cell behavior and regenerative response.
